These strains produced unconjugated asci with one or two ellipsoidal to elongate ascospores in in a persistent ascus. Phylogenetic analysis of the D1/D2 domains of the large subunit (LSU) rRNA gene sequences indicated that the isolates represent two novel sexual species in the Candida/Lodderomyces clade. Candida baotianmanensis sp. nov. was located in a statistically well-supported branch together with Candida maltosa. Candida pseudoviswanathii sp. nov. formed a subclade with its closest relative Candida viswanathii supported by a strong bootstrap value. The two new species were distinguished from their closest described species, C. maltosa and C. viswanathii, in the D1/D2 LSU rRNA gene and ITS sequences and in phenotypic traits. The type strain of Candida baotianmanensis sp. nov. is NYNU 14719T (= CICC 33052 T= CBS 13915 T), and the type strain of Candida pseudoviswanathii sp. nov. is NYNU 14772 T (= CICC 33053T= CBS 13916 T). We present 7 new records of laboulbenialean parasites on Staphylinidae (rove beetles) and Coccinellidae (lady beetles) from the Boston Harbor Islands National Recreation Area. These are Clonophoromyces nipponicus Terada & I.I. Tav., Hesperomyces virescens Thaxt., Ilyomyces cf. mairei F. Picard, Laboulbenia philonthi Thaxt., Peyritschiella protea Thaxt., Stichomyces conosomatis Thaxt., and Teratomyces actobii Thaxt. One of these parasite species, C. nipponicus, has not been found previously outside of its type locality in Japan. Examination of Roland Thaxter's 1891–1932 slides led to the designation of lectotypes for L. philonthi, P. protea, S. conosomatis, and T. actobii. The following synonymy is established: Teratomyces brevicaulis Thaxt. = T. actobii. In addition, we discovered new localities for H. virescens (from Canada, Cuba, Guatemala, and Japan) and L. philonthi (from Canada, Grenada, Panama, Trinidad, and Venezuela). The structures of the new compounds were elucidated by extensive spectroscopic analysis and chemical transformation. The inhibitory effects of 1–26 on HMG-CoA reductase and α-glucosidase were tested in vitro. Compounds 1, 3, 6, 10–14, 17, 18, 23, 25, and 26 showed much stronger inhibitory activity against HMG-CoA reductase than the positive control atorvastatin. Compounds 13, 14, and 16 presented potent inhibitory activity against α-glucosidase from yeast with IC50 values of 13.6, 2.5, and 5.9 μM, respectively. In addition, the cytotoxicity of 1–26 was evaluated against the K562 and PC-3 cell lines by the MTT assay. Compounds 1, 2, 6, 7, 10, 12, 16, 18, and 25 exhibited cytotoxicity against K562 cells with IC50 values in the range 10–20 μM. Paclitaxel was used as the positive control with an IC50 value of 0.9 μM. This is the first report of secondary metabolites from this medicinal mushroom. Here we evaluated the pre-penetration processes in leaves at different phenological stages of a susceptible clone (3918) of Eucalyptus grandis. The first ten leaves, from the tip to the base, on the apical branch were detached and inoculated with urediniospores. We found a gradual reduction in fungal germination (88 to 39 %), apressorium formation (78 to 18 %), penetration (69 to 0 %) and sporulation (90 to 0 number of sorus /cm2) with the increase in leaf age. In addition, thickness and amount of wax was higher in the older leaves. The results of this study suggest that resistance to P. psidii infection in older Eucalyptus leaves occurs at the pre-penetration stage. Their ecological versatility could imply a capacity to produce diverse secondary metabolites, but only a few studies have aimed at characterizing their chemical profiles. Here, we assessed the secondary metabolites produced by five Exophiala sp. strains of a particular phylotype, isolated from roots of Microthlaspi perfoliatum growing in different European localities. Exophillic acid and two previously undescribed compounds were isolated from these strains, and their structures were elucidated by spectroscopic methods using MS, 1D and 2D NMR. Bioassays revealed a weak activity of these compounds against disease-causing protozoa and mammalian cells. In addition, 18 related structures were identified by UPLC/MS based on comparisons with the isolated structures. Three Exophiala strains produced derivatives containing a β-d-glucopyranoside moiety, and their colony morphology was distinct from the other two strains, which produced derivatives lacking β-d-glucopyranoside. Whether the chemical/morphological strain types represent variants of the same genotype or independent genetic populations within Exophiala remains to be evaluated. ITS and D1/D2 sequencing was employed to further identify the organism as Chrysosporium guarroi. Based on sensitivy results from the mycotic isolate collected from the patient, ketaconozole therapy was initiated along with topical povidone-iodine. The iguana was re-evaluated after a month and new healthy skin was observed at the affected area. The habitat, origin, transmission, treatment and the primary or secondary role of Chrysosporium spp. are unknown in iguanas, as well as the zoonotic nature of this microbe. This report describes a pet green iguana diagnosed with cutaneous mycosis caused by Chrysosporium guarroi in Turkey. As there is relatively little published information describing Chrysosporium guarroi in reptiles, further studies are required to understand the importance of this pathogen in captive and wild animals. We isolated five secondary metabolites with a p-terphenyl backbone from the mycelial extract of a fermentation of this fungus in 70 l scale by using RP-HPLC, which were named rickenyls A–E (1–5). Their structures were elucidated by X-ray crystallography and NMR spectroscopy, complemented by HRESIMS. Two of the compounds contained a quinone core structure in ortho (2) and para-position (5), respectively. We obtained 2 spontaneously and by lead tetraacetate oxidation from 1. All compounds were screened for antimicrobial, antioxidative and cytotoxic activities. Rickenyl A (1) exhibited strong antioxidative effects and moderate cytotoxic activity against various cancer cell lines. The current report focuses on the diagnostic feasibility of different methods, paying particular emphasis on the application of the Mini-FLOTAC technique for diagnosis of M. ornithogaster. Mini-FLOTAC is particularly tailored for epidemiological monitoring and surveillance, where large numbers of faecal samples must be rapidly, yet reliably examined. Gram stain as standard method was used to validate the reliability of Mini-FLOTAC. This tool has not yet been used in avian species nor even in the diagnosis of yeast infections. In our study M. ornithogaster showed an excellent flotation performance never proved. Our results suggest that Mini-FLOTAC is a valid, sensitive and potentially low-cost alternative technique to use in the diagnosis of this yeast infections in birds.
